The mission of the engineer will be to develop existing partnerships and new industrial partnerships, on a national and European scale for the ChemBioFrance (CBF) research infrastructure. More specifically, he/she will: - establish contacts between biopharma companies and the ChemBioFrance research infrastructure , in order to set up partnerships ensuring the sustainability of the National Chemical Library and the maintenance at the highest level, equipments for screening and development of molecules in the field non-regulatory preclinical drug discovery and development. - support the director and the members of CBF executive committee in contacts with the private sector to present services, organize negotiations and prepare contractual documents in interaction with ChemBioFrance supervisory authorities. Activities of the business developper are : - establish contacts with french SATTs (technology transfer assistance companies) in France to present ChemBioFrance services and take part to prematuration and maturation programs - contact all french start-ups working in the field of drug development, and obtain requests for quotes and services - for these same start-ups, present the partnership offer and negotiate contracts for nataional chemical library co-development. Discuss the opportunity to set up LabComs to help them develop their offer - allow the CBF executive committee to apply to European programs - develop and coordinate a network of key industrialists in connection with the themes of the CBF laboratory users - maintain a Customer Relationship Management - ensure forecast and follow-up related to its activity - edit activity reports - develop communication support - conduct satisfaction surveys.

L'ingénieur(e) aura pour mission de mettre en œuvre dans un premier temps un Plan de Gestion de Données (PGD), qui consiste à synthétiser la description et l'évolution des données durant un projet de recherche. Dans un second temps, il/elle devra construire, alimenter les bases de données, en les rendant accessibles et pérennes dans le contexte de l'Open Data (processus de FAIRisation). Il/elle administrera également les bases de données ainsi que leurs accès web et le parc informatique de l'unité. Il/elle participera à la politique qualité de l'unité.

L'assistant ingénieur aura à réaliser, en s'adaptant aux conditions d'expériences, un ensemble de techniques spécialisées de préparation, d'analyse, de caractérisation d'échantillons et dans un second temps, de criblages robotisés haut débit. In the frame of a collaborative project aiming at discovering new binders for neurodegenerative diseases, our group is developing new labelling technologies, combinatorial chemistry and high throughput screening for application in nuclear imaging (see Angew. Chem. Int. Ed. 2018, 57, 9744; Chem. Commun. 2021, DOI: 10.1039/D1CC02665H). The project aims to develop new combinatorial synthetic approaches to prepare libraries of molecules which will be used to accelerate the discovery of new ligands able to selectively bind protein aggregates involved in Alzheimer, Parkinson and Huntington diseases. The strategy is based on Click Chemistry combined with a mass enhancer and radiolabeling, to identify specific ligands for neurodegenerative diseases and map their distribution by imaging. The postdoctoral fellow will be hosted in the SCBM (Service de Chimie Bioorganique et de Marquage) at CEA-Saclay and will work in close collaboration with several teams within the network. All experiments will be conducted with stable isotopes. CEA personnel will perform experiments with radiolabeled compounds.

Le stagiaire en master 2/ingénieur informatique aura pour mission de participer à la FAIRisation des données générées par l'Infrastructure de Recherche ChemBioFrance : - Faciles à retrouver (moteur de recherche, identifiants pérennes pour les données et les méta-données) - Accessibles (via https par ex.) - Interopérables (exportables vers un fichier csv par ex.) - Réutilisables (les conditions d'utilisation des données doivent être clairement précisées). Dans un premier temps il/elle travaillera à la « curation » des jeux de données existants sous forme de fichier Excel (plus de 100 jeux issus des criblages des composés de la Chimiothèque Nationale). Il/elle proposera ensuite un cahier des charges qui inclura la validation d'un schéma de données s'inspirant largement de la base de ChemBL, ainsi qu'une description fonctionnelle des besoins. Il/elle implémentera la base de données et l'alimentera, en la rendant accessible et interfaçable, dans le contexte de l'Open Data.

In the frame of a collaborative project aiming at discovering new scaffolds of interest, our groups are developing new synthesis methodologies, using innovative technologies to accelerate the optimization and scope processes. This project aims at developing catalytic enantioselective cycloadditions of sydnones in order to efficiently access chiral pyrrazolines. The design of the corresponding catalytic process will rely on the concept of dual catalysis in which both the dipolarophile and the sydnone could be activated. We will study cycloaddition reactions with either electron rich or electron-poor dipolarophiles, which may be activated with a chiral catalyst, while the sydnone may be activated by energy transfer via a photocatalyst from visible light. A High Throughput Experimentation (HTE) approach will be deployed to empower and speed-up the discovery and the optimization of suitable catalytic conditions, and exploring a large number of dipolarophiles.

In the frame of a collaborative project aiming at discovering compounds that interfere with the intracellular retrograde trafficking, our group is developing small molecules that have been shown by us and other groups to have a broad "therapeutic" activity (look for Retro-2 references) . Based on the experience we acquired on Retro-2 analogs (see for instance J. Med. Chem. 2013, 56, 8, 3404-3413) and based on in silico design we now aim at developing a completely new chemical series for which we already have encouraging preliminary results. The postdoctoral fellow will be hosted at the SCBM (Service de Chimie Bioorganique et de Marquage/Université Paris-Saclay) at CEA-Saclay under the supervision of Dr Jean-Christophe Cintrat and will work in close collaboration with biology team within the network. All experiments will be dealing with heterocyclic chemistry and may require some basic knowledge of in silico drug design.

Basophils are rare immune cells expressed in the human body. They are involved in several biological functions, such as defence against helminth parasite and connection between the innate and adaptive immune compartments. However, because of their rapid activation in reaction to a wide range of stimuli, they are directly involved in pathogenic processes such as atopic/allergic and inflammatory processes, in both respiratory and skin systems (asthma, rhinitis, urticaria, etc¿). It has been shown that the IL3 cytokine is a key protein in the activation of basophils processes and inflammatory response. Thus, the candidate will study IL3 cytokine in order to develop small molecules targeting the protein. Bioinformatics analysis, molecular dynamics simulations and drug design approaches will be used for the design of inhibitors of the IL3 pathway, in order to suppress basophil activation under pathological conditions. The project is financially supported by the ANR (Agence Nationale pour la Recherche, BASIN project, ANR-19-CE17-0021) and involves collaboration with several experimental labs.

A post-doctoral position is currently available in the Cellular and Chemical Biology unit (U1143 INSERM ¿ UMR3666 CNRS; http://minilien.curie.fr/89q4ev), in the Endocytic Trafficking and Intracellular Delivery team of Dr. Ludger Johannes, in tight collaboration with the Chemical Biology of Cancer team of Dr. Raphaël Rodriguez. The candidate will test different approaches to conjugate chemical compounds with immunomodulatory properties to STxB. The program will begin with small molecules; upon success, other entities such as nucleic acids will be designed to be grafted to STxB.
